The Punjab government on Thursday approved the modalities for distribution of smart mobile phones to the State’s youth, to pave the way for the implementation of its ‘Mobile Phones to the Youth’ scheme. The decision was taken at a Cabinet meeting presided by Chief Minister Captain Amarinder Singh at Dera Baba Nanak. “The vendor to implement the scheme would be selected through an open transparent bidding process,” said an official statement, adding that the first batch of phones is expected to be distributed in December 2019In the first phase, the mobile phones would be distributed to girl students in Class 11 and 12 of government schools in the State. The State government had announced its ‘Mobile Phones to the Youth’ scheme, in line with its poll promise, in its Budget for the financial year 2017-18, and the due allocation of funds was made in the Budget for 2018-19.“The scheme is aimed at providing digital access to youth and information regarding education, career opportunities, access to skill development and employment opportunities, access to basic citizen services through government applications etc,” said the statement.

Kolkata: The Election Commission has served a show-cause notice each to the BJP district president of Nadia Mahadev Sarkar and Trinamool Congress MLA from Chakdah Ratna Ghosh Kar, for their derogatory remarks. But what if the standards for what makes a good or bad performance had evolved as leaguewide numbers changed? Pro-Football-Reference.com does a great job of adjusting for era with its Advanced Passing indices, which are centered on an average of 100 with 15 points representing 1 standard deviation in either direction. But I wanted to rescale the building blocks of passer rating itself to see how today’s passing numbers would translate to a rating if the NFL had simply allowed its rating system to change with the times.To do that, I looked at the distribution of stats in each category that goes into passer rating — completion percentage, yards per attempt, touchdown percentage and interception rate — from the sample originally used to craft the formula back in the early 1970s (qualified passers from 1960 to 1970). Specifically, I figured out the spread of values (relative to the league) that, in a given category, led to the minimum number of points (0), the average number of points (1) and the maximum (2.375). Under the hood, passer rating is built around these ranges; it hands out points on that 0-to-2.375 scale in each category, then sums up the four values, divides by 6 and multiplies by 100. (Hence, 67 is supposed to be average — a 1.0 in four categories, divided by 6, times 100.)For any era, we can rescale what performance “should” lead to a given value in each category to keep the relative leaguewide distribution the same as it was when passer rating was first conceived.2Just like with PFR’s advanced passing stats, I calculated these distributions using qualified passers in rolling three-season periods to avoid a strange spread in one season causing overly volatile results. (To qualify, a passer needed at least 14 attempts per team game.) So while, say, Alex Smith’s 62.5 percent completion rate in 2018 was worth 1.0 point, so was Don Meredith’s 49.5 percent mark from 1962. Do this for every category in every season, and you have a stabilized version of passer rating that no longer spirals uncontrollably upward with each innovation in the passing game.Some ultra-high ratings change less than you might expect under this new method. Rodgers’s single-season record of 122.5 from 2011 tumbles all the way down to … 121.1. (He was very good that year.) But other seemingly immortal ratings, such as Kirk Cousins’s 99.7 mark this season, get knocked down quite a bit — in Cousins’s case, he falls to a much more reasonable 81.5 rating. But passer rating itself has always been a surprisingly decent metric within any self-contained era; the team with the higher passer rating (by any margin) in a game wins about 80 percent of the time. It’s the comparisons across eras that have become distorted as the game has changed over time. But a simple fix tethering modern stats to the standards contained in passer rating’s formula would go a long way toward restoring sanity to the metric you still see in every NFL box score and broadcast. Now, it’s four straight losses, an increased risk of dropping out of the top 25 and a season spiraling out of control.The Ohio State men’s basketball team has had an odd feeling about it lately.“It’s definitely been different. No one in that locker room has ever lost like this,” junior forward Sam Thompson said. “No one has ever lost four in a row and we know that it’s unacceptable for our program to lose four basketball games in a row.”The current skid is the first time OSU has lost four straight games since February 2009. In those four losses — to Michigan State, Iowa, Minnesota and Monday at Nebraska — the Buckeyes (15-4, 2-4) have struggled shooting the ball, making just 40.4 percent of their shots from the field. Points in the paint have been an issue as well, particularly in the last three games, as OSU has given up 120 points down low.Junior centers Amir Williams and Trey McDonald have struggled, causing OSU coach Thad Matta to play with a smaller lineup down the stretch late in games to give his team a chance. Williams has started all 19 games this season for OSU — along with senior guards Aaron Craft and Lenzelle Smith Jr., junior guard Shannon Scott and junior forward LaQuinton Ross — but his recent play begs a question: Should there be a change to the starting five?“I haven’t even thought in terms of that, from what we did (Tuesday) to what we’re getting to go out and do here in practice. We’re going to compete and get after it and I want to see who’s standing at the end of the day, but I’ve had a lot of thoughts go through my mind. But the starting lineup — I haven’t even thought about that one,” Matta said after a brief pause. “We’ve started games well. Last couple, we’ve got off to decent starts. I’ve been pretty pleased with those, but we gotta get the middle and the end corrected.”No matter who starts or finishes the games, though, the end result has not been what OSU wants, as turnovers, missed shots and poor defensive executions have led to almost three weeks without a victory.“I don’t know if I’d call it a lack of confidence. I’d definitely say there is a different feel around than we had (19) days ago, but I wouldn’t call it a confidence issue,” Thompson said. “I would say there is a certain frustration level with the basketball team, just with losing. Tom Herman, now the University of Texas head coach and former offensive coordinator under Meyer, was named by Snook as the person who told former ESPN reporter Brett McMurphy about Meyer’s alleged knowledge of domestic abuse from Zach Smith.Snook’s report states that Herman was “furious” after 2019 five-star receiver Garrett Wilson committed to the Buckeyes in April, which, according to the report, was his motive behind the leaked information. According to the report, while at Ohio State, Herman encouraged Smith’s now ex-wife Courtney Smith to “go public” with the alleged domestic violence incidents while he was a member of the Ohio State staff. -Around 6000 professionals have been working round the clock to complete construction work on the 1,320MW coal-fired Payra power plant at Dhankhali in Patuakhali’s Kalapara upazila.Being built by Bangladesh-China Power Company Ltd (BCPCL), the plant, if completed, would be country’s largest coal-fired power plant.The BCPCL is a 50:50 joint venture between Bangladesh’s North-West Power Generation Company Limited (NWPGCL) and China National Machinery Import and Export Corporation (CMC).During a visit to the project on Friday, around 1,500 Chinese and some 4,500 local workers were seen busy working on the completion of the project on time.Around 35 per cent of the construction work of the project, including piling for infrastructure building, constructing an iron frame for first unit’s boiler, project roads, and more is already complete, said BCPCL officials.They expected they would be able to bring in the required machineries for the plant in the first half of 2018.The estimated cost of the project is $1.6 billion, 80 per cent of which is expected to come as a loan from China’s Exim Bank. The rest would be invested by NWPGCL and CMC.The EPC contractor, which won the contract, is a consortium of First Northeast Electric Power Engineering Company (NEPC) of China and China National Energy Engineering and Construction Company Ltd (CECC).NWPGCL managing director AM Khurshedul Alam told Prothom Alo that there would be two 660MW units in the project.The first unit of the power plant is expected to supply electricity by April 2019 and the second unit will start generation in October, he added.The NWPGCL has been allotted land of 1000 acres near Payra port. William Barber II, a progressive activist and pastor, addressed the executive committee of the Democratic National Committee on Friday (Aug. 23), calling on party leaders to host a presidential debate focused on poverty and to do more to address the concerns of poor and low-income Americans.The co-chair of the Poor People’s Campaign, named for the last major campaign of Martin Luther King Jr., Barber insisted that poverty touches all people and regions of the United States and overlaps with related concerns such as racism, voter suppression, healthcare, “ecological justice,” militarism and religious nationalism.“We have to say the word, ‘Poverty,’” said Barber, according to a copy of his remarks provided to Religion News Service. “We need to lift up the stories of folks in Appalachia and Kansas and the Mississippi Delta. On Thursday (Aug. 22), the DNC resolutions committee voted 17-8 against holding a debate focused on climate change, sparking outrage among environmental activists.Asked about a potential poverty debate at a candidate forum hosted by the Poor People’s Campaign in June, however, all nine Democratic presidential hopefuls who attended — including former Vice President Joe Biden and Sens. Elizabeth Warren, Bernie Sanders, and Kamala Harris — said they would support it.The Poor People’s Campaign, which now has established chapters in more than 40 states, has long championed the notion of creating a “new electorate” of poor and low-income Americans, who are often less likely to vote than other Americans. If mobilized, Barber and his allies argue, the less well-off could potentially swing elections.In his address, Barber announced his intention to launch a nine-month, 22-stop “We Will Do MORE” campaign focused on mobilizing, organizing, registering and educating poorer Americans. In addition, Barber argued in his speech that the issue of poverty is not about “left versus right, but right versus wrong.”Even so, the pastor, who helped lead North Carolina’s Moral Mondays protests credited with helping to unseat the state’s Republican governor in 2016 — had harsh words for Republican Senate Majority Leader Mitch McConnell, comparing his decision to block efforts to pass voting rights legislation to infamous segregationist Strom Thurmond’s decision to filibuster the Civil Rights Act in 1957.Barber also voiced thinly veiled criticism of the Trump administration’s policies and of Christians who support them.“For too long, faith has been hijacked by those who say abortion and sexuality are the only moral issues,” Barber said. “They tell religious people to vote their pro-life/pro-family values, then they use their political power to pass policies that keep families in poverty, separate families at the border, keep families from getting healthcare and endanger the lives of children in schools, worshipers in churches and the planet itself.”He invoked scripture to dismiss claims that polices designed to help the poor would be tantamount to socialism.“If someone calls it socialism, then we must compel them to acknowledge that the Bible promotes socialism,” he said. “This current administration is practicing socialism to corporations and the greedy through tax cuts, deregulation and economic incentives. By Stephen Janis, Special to the AFROAs funeral services for slain Baltimore Homicide detective Sean Suiter were held this week, the mystery has only deepened about how the veteran cop was killed in a vacant West Baltimore lot roughly two weeks ago.There have been few leads and even fewer clues as to how and why Det. Suiter, an 18 year veteran of the Baltimore Police Department (BPD), was fatally shot in the head with his own gun after what police have described as a brief struggle.Slain Baltimore Homicide detective Sean Suiter. (Courtesy Photo)But, one fact which has emerged from the case is only stoking community suspicions and raising more questions about the department tasked with solving the case: the widening of an investigation into eight officers charged with stealing from residents, racketeering and drug dealing.At an impromptu press conference on Nov. 22, BPD Commissioner Kevin Davis revealed that Suiter was set to testify before a federal grand jury as part of an ongoing investigation into the now-notorious Gun Trace Task Force. The charges brought against that group have continued to reverberate throughout a department already under a federal consent decree and battling a record wave of crime and homicide.“I am now aware of Det. Suiter’s pending federal grand jury testimony surrounding an incident that occurred seven years ago with BPD police officers who were federally indicted,” the commissioner said. “The acting U.S. attorney and the special agent in charge of the Baltimore field office have told me in no uncertain terms [that] Detective Suiter was not the target of any ongoing criminal investigation.”The bombshell announcement came about a week after Suiter was killed.But, tangible leads in the slaying have been scarce, according to investigators familiar with the case. That fact has not stopped Davis from publicly expressing his theories on what happened—among them, that Suiter’s impending testimony and his murder are not linked.“It appears to be nothing more than a spontaneous observation of man acting suspiciously, and a spontaneous decision to investigate his conduct,” Davis said.Still, the fact that Davis’ comments came during the widening ongoing federal investigation of the department, and during a high-profile murder investigation, have raised questions both inside and outside the department.“Everyone was surprised that he’s been talking so freely about the case,” a source familiar with the investigation, who did not want to be named, told the AFRO.Suiter was not the only officer who worked with the Task Force to receive a summons to appear before a federal grand jury. According to sources within BPD, at least one other officer who had worked with the Gun Trace Task Force received a letter last week compelling them to testify, indicating federal prosecutors are far from done with the scandal.It’s unclear which aspect of the case investigators are currently exploring. However, Baltimore NBC affiliate WBAL-TV and The Baltimore Sun reported that federal prosecutors have reopened an accident case that involved Suiter and Sgt. Wayne Jenkins, one of the eight Task Force officers already indicted. In 2010, the pair engaged in a car chase with a suspect who ultimately crashed into another vehicle, killing the 86-year old father of a Baltimore police officer. The suspect was charged with possession of heroin, but has asked a federal judge to reopen his case.Meanwhile, questions still surround the few details police have disclosed about what happened in that vacant lot in the city’s Harlem Park neighborhood, a crime scene that remains cordoned off. An aspect of the case that troubled one former homicide investigator is how a suspect could take Suiter’s weapon and shoot him while his partner was nearby.“We would cover someone,” said former Baltimore Homicide Lt. Stephen Tabeling. “I would have been right behind him.”However, Davis has said that the detective who accompanied Suiter that day, who has yet to be identified, was allegedly not in the immediate vicinity when the shooting occurred.“Upon the sound of gunfire, Suiter’s partner sought cover across the street and immediately called 911,” Davis said. “We know this because it was captured on private surveillance video that we’ve recovered.”“The evidence refutes the notion that Detective Suiter’s partner was anything but just that, his partner,” Davis said.Tabeling also noted that Baltimore police officers usually wear safety holsters, which are designed to prevent suspects from taking a gun during close contact.“They make it pretty tough to get the gun,” Tabeling said.

All I really did was turn some screws on a plate, and I stand on the shoulders of a lot of great people in this community that really did this,” Ward told the group of Democrats, who honored him at their monthly meeting.Ward, who accepted a symbolic $1 to do the modification job, also said he was inspired to action by growing up in the era when schools were integrated.State law generally prohibits alterations to monuments without state approval. That law was enacted to protect monuments during debate over removing the Confederate flag from atop the Statehouse.A judge ruled in favor of the veterans group seeking to have the listing on the monument changed. However, the issue was still being litigated when Ward took action.The judge noted the monument was privately owned by the American Legion, even though it sits on public property. The judge said private ownership allows for changes despite the state law known as the Heritage Act.State officials initially asked the judge to reconsider because they think the decision could leave other monuments unprotected. After the monument was changed, the defendants asked the judge to vacate his ruling, saying that after the plaque switch the issue is moot.“Simply put, there now is nothing left for the Court to do and nothing left for the Court to enforce,” the defendants’ attorneys wrote in a motion to vacate a ruling. “The plaques were replaced by a person not a party to this case and, thus, the case was mooted by a non-party without regard to the actual legal issues pending in this case.”
Journal information: Proceedings of the National Academy of Sciences (Phys.org) — In a tale worthy of Sherlock Holmes, scientists in the School of Chemistry at the University of Bristol, UK have solved a biochemical mystery that had previously proven elusive for 70 years: How the fungus Talaromyces stipitatus produces stipitatic acid (6), which is a tropolone, one of an atypical group of fungal natural products – that is, small molecules produced by genetically encoded pathways – with a seven-carbon ring. (Most natural products, such as cholesterol or phenylalanine, have five or six carbons in rings.) The researchers used a two-part biosynthetic approach – gene deletion and alternate genetic expression – to investigate the molecular pathway in question. Professor Russell J. Cox, Postgraduate student Jack Davison and other researchers engaged in the study faced several long-standing obstacles to showing that 3-methylorcinaldehyde is the direct product of a fungal nonreducing polyketide synthase (NR-PKS) which most likely appends the methyl group from S-adenosyl methionine during biosynthesis of the tetraketide, and which uses a reductive release mechanism to produce the observed aldehyde. “Gene knockouts have been one of the most useful tools in the toolkit of the biosynthetic chemist,” Cox tells Phys.org, “but knockouts can’t answer complex questions like these. We now make extensive use of heterologous expression – that is, moving the gene to a ‘clean’ host and switching it on.”The scientists then monitor the host organism for the production of new compounds which we isolate and identify. The chemical structure of the new compound tells them a great deal about the chemistry which must have been used to make it. “In this case,” Cox continues, “we showed that the TropA gene encodes a polyketide synthase which makes 3-methylorcinaldehyde. Expression also allows us to do more complex experiments – for example, by truncating or mutating the gene – and in this way we discovered the reductive release mechanism and the fact that the programmed methylation occurs during chain extension rather than after chain-building and ring formation.”Tropolone biosynthesis was one of the longest-standing problems in the field of biosynthesis and some of the most distinguished organic chemists of the last century were fascinated by it, but progress had been very limited. “We realized that combining chemical knowledge with genome sequence data could give us the start we needed,” Cox explains. “We already knew a lot about polyketide biosynthesis in fungi, and this allowed us to narrow down the potential genes involved to just four. We then used chemical knowledge to narrow this further to a single gene cluster.” Proof then came from the knockout and expression studies. In collaboration with our colleagues in the School of Biological Sciences at Bristol we’re developing systems which can express a dozen or so fungal genes in parallel. This will allow the researchers to investigate much more complex systems in the future.In addition, Cox adds, “I do believe that computational biology and chemistry will eventually provide answers to complex questions like this – but at the moment, while computational methods allow us to formulate good questions, lab work is still needed to find the answers. We’re working in an area with very many unknowns, so it’s difficult for computational methods which rely on current knowledge to act predictively with any accuracy. In fact,” notes Cox, “this is a powerful reason why fundamental discoveries are still so important: they’ll form the basis for future predictions.”Cox points out that he and his team will also continue to study the TropB-D enzymes in vitro. “Chemical methods of classical enzymology will allow us to determine their precise mechanisms.”Cox also articulates how comparing the T. stipitatus tropolone biosynthetic cluster with other known gene clusters allows clarification of important steps during the biosynthesis of other fungal compounds, including the xenovulenes, citrinin, sepedonin, sclerotiorin, and asperfuranone. “Fungal genomes generally are much bigger than their bacterial counterparts by roughly 10 times, and they contain many more genes and gene clusters encoding the biosynthesis of complex compounds” he explains. “Barely any of the known fungal gene clusters have been linked to the molecules they must encode. Our work now allows the understanding of a set of genes which encodes the biosynthesis of polyketides followed by oxidative modifications and these occur frequently in fungi. Until now these clusters were mysterious, but now we can – at least partially – begin to understand what they may do. Secondly, knowledge of the gene clusters will allow us to go hunting for new clusters more effectively.” For example, puberulic acid is a potent antimalarial compound, but its gene cluster is unknown – and the team predicts that the cluster should be very similar to the T. stipitatus tropolone gene cluster.In terms of other research and applications that might benefit from their findings, Cox says that understanding biosynthetic pathways is a key strand of the new science of Synthetic Biology. “One can think of the gene clusters and biosynthetic enzymes they encode as building blocks of new biological entities. In the future,” he concludes, “it will be possible to combine the genetic and chemical knowledge of biosynthetic pathways to produce bioactive compounds – such as drugs and agrochemicals – using biology rather than chemistry. So far, backers have pledged only about $1,400. James PurnellThe BBC has called for on-demand players like its iPlayer service to be guaranteed prominence on all major TV platforms.In a article for the Telegraph ahead of a House of Lords debate on the issue this week, the BBC’s director, radio and education, James Purnell, said it is important that content from the BBC and other public service broadcasters can be easily found in an increasingly digital age.“Fourteen years ago, Parliament took a far-sighted decision. It insisted that the public service channels including BBC One, ITV and Channel 4 should be at the top of the programme guides on all TV platforms,” said Purnell.“It made our channels easy to find – especially important for the BBC, whose programmes people have already paid for via their licence fee. But the world has changed, with the rapid growth of digital channels and new services.”He said that the new generation of set-top boxes have a limited number of content options on their home pages and that if those places are filled by content from the platform owners like Sky, or from Netflix, Amazon or YouTube, “that leaves little room for the on-demand services from our public service broadcasters”.“Some pay-TV platforms are already making ‘free to air’ services harder to find. On the new Sky box, Sky Q, there is no one button on the remote control that takes you to live TV, the single most popular thing Sky customers do. “We are on the cusp of curing aging.” Those are the words of molecular biologist Dr. William Andrews, founder and CEO of Sierra Sciences, a company with the stated goal to “cure aging or die trying.” In the quote above, which comes from a September 2010 Sierra Sciences press release, Andrews is referring to a nutraceutical (i.e., a natural food-derived product meant to provide health or medical benefits, like vitamin C or folic acid) that his company discovered, known as TA-65. It supposedly slows down the aging process in humans by activating an enzyme called telomerase. Telomerase acts on telomeres—simple repeating sequences of nucleotides (the building blocks of DNA) found at the tips of chromosomes that are important to cell division and replication. Cells must replicate their genes accurately and completely whenever they divide, or the so-called daughter cell will malfunction and die. However, most DNA polymerases (the enzymes that replicate DNA) cannot copy chromosomes all the way to the end. Thus, during each cell division, a small region at the tip of the chromosome (part of the telomere) remains uncopied. So, the telomeres have a crucial function: they act as a buffer zone during cell division, protecting the genes in the chromosome and ensuring that they fully replicate. But there’s a cost: successive divisions cause the telomeres to erode over time. When telomeres erode away, parts of critical genes remain uncopied, eventually spelling death for the cell. Some scientists think that this telomere erosion is what imposes a limit on our lifespans and causes our health to decline as we age; they also think that an answer to this dilemma might be telomerase activation. Telomerase combats telomere erosion by adding telomeric subunits (repeating sequences of nucleotides) to the ends of the chromosome, thereby allowing it to continue to divide intact. The origin of telomerase still remains a mystery. It appears to be critical during human embryonic development, but then fades away. Little is found in the body after birth, for reasons we don’t yet understand. Some scientists, like Dr. Andrews, think telomerase activation could be the cure for aging that his company is looking for, and he views TA-65 as a potential big first step in that direction. According to Andrews: TA-65 is going to go down in history as the first supplement you can take that doesn’t merely extend your life a few years by improving your health, but actually affects the underlying mechanisms of aging. Better telomerase inducers will be developed in the coming years, but TA-65 is the first of a whole new family of telomerase-activating therapies that could eventually keep us young and healthy forever. Well, if proven true, that’s a game-changer, to say the least. Just keep on living until you meet with a fatal accident. Woo hoo! But… hold on. Turns out we shouldn’t get irrationally exuberant just yet. It might not be such a good idea to activate telomerase, after all—because of the role it seems to play in cancer. Cancers can arise when genetic mutations inside a cell cause it to escape from normal controls on replication and migration. The cell and its offspring multiply uncontrollably while invading and damaging nearby tissue. With the presence of telomerase, cancerous cells—which apparently synthesize the enzyme—avoid telomere erosion. They essentially become immortal, dividing for as long as the host survives. A 2012 study by the MD Anderson Cancer Center found that activating telomerase following telomere degradation actually made tumors stronger and more deadly. So alas, TA-65 and telomerase activation might not be the miracle aging “cure” Dr. Andrews was hoping for. But what about other areas of anti-aging/longevity research? There’s a lot of talk these days about humans becoming essentially immortal in the near future, but is the hype supported by any actual research? Let’s see. The Quest for Immortality Continues People have been obsessed with the idea of immortality and living forever for centuries. According to Adam Gollner in The Book of Immortality: The Science, Belief, and Magic Behind Living Forever: The twenty-five-year old Emperor Ai of Jin died in 365 CE, after overdosing on longevity drugs. He wasn’t the last leader to die trying to live forever. The fascination with chemical immortality reached an ironic apogee centuries later, during the T’ang dynasty (618-907 CE), when elixirs poisoned those hoping for precisely the opposite effect. In the late 1300s, legend has it that alchemist Nicolas Flamel created a “sorcerer’s stone” that was then used to produce a potion, the elixir of life, which is said to make the drinker immortal. The idea was so alluring that despite a complete lack of evidence to support the existence of the stone and the elixir, other scientists, including the great Sir Isaac Newton, later tried to replicate Flamel’s results. To no avail. And of course there’s the fabled Fountain of Youth that became associated with Spanish explorer Ponce de León in the early 1500s. We know what happened to him. People today are no different. Not wanting to die is as natural as life itself. Below we’ll present a few interesting discoveries that may (or may not) mark some progress in our quest to live forever. But first we must answer a question. What Is Anti-Aging Medicine? The telomere erosion we talked about earlier certainly plays a part in aging, but most scientists don’t believe that it’s the only factor. They typically view aging as a sort of ongoing cellular wear and tear, where stresses from the environment and inside our cells (such as errors in DNA replication) accumulate throughout life and eventually wear out our cells and tissues. Scientists who search for a modern anti-aging elixir seek to slow down or reverse this process in order to extend both the maximum and average lifespan of people. And even if that doesn’t pan out, advocates of anti-aging medicine contend that at the least, targeting and altering the underlying mechanisms of aging will bring us more efficient ways to deal with age-related conditions like heart disease and many cancers. Now critics dispute the portrayal of aging itself as a disease, of course, but that’s not for us to debate here. Let’s just agree with the commonly stated goal of anti-aging medicine, to “add more years to your life, and life to your years.” Caloric Restriction Mimetics In 1934, scientists at Cornell University found that mice on an abnormally low-calorie diet lived about twice as long as mice who ate as much as they wanted. Scientists have since found that this caloric restriction also lengthens the lives of fruit flies, rats, and even primates, suggesting that this is an avenue worth pursuing in anti-aging medicine. The going (very basic) hypothesis of why these animals live longer is that their bodies treat food scarcity as an extreme type of stress. So they mount a physiological response to cope with the lack of nutrition. And that toughens them up, promoting health and longevity during the time of deprivation, even if that time is significantly extended. Starving yourself in order to get stronger? That sure seems counterintuitive. But the results from numerous experiments make a compelling case. The problem with caloric restriction is that it’s not a very pleasant way to live if you’re fortunate enough to be able to consume a “normal” amount of food on a regular basis (which is between 2,400 and 3,000 calories a day for active males). So scientists are trying to develop drugs (called caloric restriction mimetic drugs) that re-create the anti-aging benefits of caloric restriction without having to change one’s diet. Resveratrol, which is found in the skin of red grapes, was an early favorite in this category. It was first thought to activate a class of enzymes called sirtuins that have been associated with the anti-aging benefits of caloric restriction; but more recent studies have raised serious doubts about whether this compound (and more potent versions of it) actually activates these enzymes and has life-extending benefits. A drug called rapamycin has shown more promise as a caloric restriction mimetic by inhibiting a molecular signaling pathway called TOR. The TOR pathway acts as sort of as a food sensor and helps regulate the body’s response to nutrient availability. Blocking it has been shown on multiple occasions to extend the lifespan of lab knockout mice while keeping them lean and healthy in their later years. Unfortunately, however, the drug’s anti-aging potential in humans is offset by its potent immunosuppressant effects. Overall, research in this field has a long way to go. NAD As we noted above, aging is a sort of cellular wear and tear. According to Dr. David Sinclair of Harvard Medical School, there’s a specific biological signal that accompanies aging and tells cells it’s time to check out. It’s triggered when the cell perceives a lack of oxygen. That makes the mitochondria less efficient at converting fuel (such as glucose) into the ATP needed for cells to function properly. But Sinclair and his colleagues recently found a way to counteract that signal. Using a natural chemical compound called NAD, they were able to revive older cells in mice and make them appear energetic and young again. After receiving NAD for just one week, two-year-old mice tissue came to resemble that of six-month-old mice. “When we give the molecule, the cells think oxygen levels are normal and everything revs back up again,” Sinclair explained. “If a body is slowly falling apart and losing the ability to regulate itself effectively, we can get it back on track to what it was in its 20s and 30s.” Let’s hope. FOXO Molecular biologist Cynthia Kenyon thinks she may have found another of the keys to a long life through her study of tiny worms called C. elegans. By tweaking just one gene in these worms (to simply make it more active), she was able to take two-week-old C. elegans (which is a creaky old age for these things) and make them appear about half that age. And her modified worms lived twice as long as the normal worms. “So they’re like 90-year-old people who look 45,” said Kenyon. The gene Kenyon changed: FOXO. FOXO is a sort of master gene that helps C. elegans protect and repair its tissues (and live longer) by controlling a number of other genes. According to Kenyon: You can think of it as a superintendent of a building. So if you have a building, a nice big building, obviously it has to be maintained. What FOXO does, or the building superintendent does, is to keep the building in good working order. It makes sure that the walls are painted, by hiring painters; it makes sure that the floors are swept. The building superintendent would hire workers to do these different things. What FOXO does, in the cell, is it switches on other genes … I’d say, altogether, there are probably about a hundred worker genes that have very important roles. And, together, what you get is a cell or tissue or an animal that stays in really good working condition for a lot longer. What’s particularly exciting about Kenyon’s work is that the FOXO gene is also found in humans, and that a more protective version of FOXO (as in Kenyon’s modified worms) is associated with longer, healthier lives. Molecular biologist Timothy Donlon found in his studies that if you have this more protective version of FOXO, you have a twofold greater chance of living to 100. And if you have two copies of it, you have a threefold greater chance of living to 100, while remaining healthy. So the gene is indeed associated with adding life to your years and years to your life. If scientists could create a drug to tweak FOXO in humans like Kenyon did in her worms, it seems like we’d have an effective anti-aging medicine on our hands. We could go on with other examples of promising research in anti-aging medicine, but that’s a reasonable sample. It’s the ultimate area of scientific research, really. Moreover, it seems likely to bring benefits to humankind (and also be lucrative for savvy investors) whether aging is a disease that can be “cured” or not. This article is included in Entrepreneur Voices on Company Culture, a new book containing insights from more than 20 contributors, entrepreneurs, and thought leaders.During a Tuesday meeting to discuss an overhaul of Uber’s company culture, former board member David Bonderman proved exactly why the conversations needed to happen in the first place.At the start of the all-hands meeting, Arianna Huffington, who was the first woman to join the company’s board last year, announced the addition of Nestlé executive Wan Ling Martello to the board, increasing the representation of women on the board from 14 to 25 percent.“There’s a lot of data that shows when there’s one woman on the board, it’s much more likely that there will be a second woman on the board,” Huffington noted.That’s when Bonderman interrupted.“Actually what it shows is it’s much likely to be more talking.”In the recording obtained by Yahoo Finance, you can hear Huffington respond at first with awkward laughter. And then her reply: “Oh. Come on, David. Don’t worry, David will have a lot more talking to do as well.”That is not to be the case, as Bonderman resigned shortly after the meeting, which, it bears repeating, was convened to talk about recommendations to change Uber’s culture after allegations of systemic discrimination and sexual harassment.Related: 3 Lessons Every CEO Can Learn From Travis Kalanick’s Emotional Meeting With Uber’s Female EngineersBonderman, the 74-year-old founding partner of investment firm TPG Capital, apologized for the “disrespectful” comment via an internal memo to Uber employees and issued this statement about his departure:”I do not want my comments to create distraction as Uber works to build a culture of which we can be proud. I need to hold myself to the same standards that we’re asking Uber to adopt. Therefore, I have decided to resign from Uber’s board of directors, effective tomorrow morning.”Earlier on Tuesday, CEO Travis Kalanick also announced that he would be taking a leave of absence in order to, as he shared in an email to the company, “take some time off of the day-to-day to grieve my mother, whom I buried on Friday, to reflect, to work on myself, and to focus on building out a world-class leadership team. The ultimate responsibility, for where we’ve gotten and how we’ve gotten here rests on my shoulders,” Kalanick wrote. “There is of course much to be proud of but there is much to improve. For Uber 2.0 to succeed there is nothing more important than dedicating my time to building out the leadership team. But if we are going to work on Uber 2.0, I also need to work on Travis 2.0 to become the leader that this company needs and that you deserve.”Related: Travis Kalanick to Take a Leave of Absence. Here Are 9 CEOs Who Were Fired or Decided to Step Down.So what exactly is Uber 2.0 going to look like?Over the past few months, former attorney general Eric Holder and Tammy Albarrán, partners at law firm Covington & Burling, were tasked with investigating the allegations made by former engineer Susan Fowler and others.They interviewed more than 200 Uber employees and provided a set of recommendations pertaining to how the company should address discrimination, harassment and retaliation and how it could “ensure that its commitment to a diverse and inclusive workplace was reflected not only in the company’s policies but made real in the experiences of each of Uber’s employees.”The recommendations include:Providing more support for the human resources department and establishing clear protocols to track complaints.Mandatory training for senior executives, HR staffers, managers and people in the position to interview prospective employees, particularly around the topics of promoting inclusion and combatting unconscious bias.In terms of recruiting and developing talent, implementing a blind resume review, and increased transparency when it comes to performance reviews and promotions.The prohibition of romantic or intimate relationships between supervisor/subordinates and the consumption of alcohol during work hours, at after work events and at company-sponsored events.An update of discrimination and harassment policies including instituting a zero-tolerance policy for violators of those rules — no matter what position they hold in the company — and explicit protection against harassment from not only other employees, but third-parties the company deals with such as clients to vendors.Related: A 2013 Letter From Uber’s Travis Kalanick Lays Out Sex Rules for Employees at Company PartyA lot of these seem quite standard. But when you look at former engineer Susan Fowler’s blog post and Kalanick’s 2013 Miami letter side by side, the necessity for Holder and Albarrán to explicitly state things such as the importance of “de-emphasizing alcohol as a component of work events,” or that policies should be applied consistently across the company with no special treatment afforded to any one employee is pretty glaring.One passage in particular about Uber’s 14 cultural values speaks volumes about where Uber ran aground. Holder and Albarrán recommended that the company communicate with its employees to reassess and develop a core list of values that are accessible and easy to understand. “Eliminate those values which have been identified as redundant or as having been used to justify poor behavior, including Let Builders Build, Always Be Hustlin’, Meritocracy and Toe-Stepping, and Principled Confrontation; and encourage senior leaders to exhibit the values on a daily basis and to model a more collaborative and inclusive Uber culture,” the recommendation reads. “Leaders who embody these values should be part of the process of redefining Uber’s values and should be role models for other leaders within the company.”Related: Uber CEO Travis Kalanick Says He Needs to ‘Grow Up’ After Video of Argument With a Driver ReleasedUber has long had a reputation for aggressive tactics in its dealings with regulators and competitors. It was seen, for better or worse, as one of the key drivers of its rapid growth. But clearly, as Uber’s experience shows us, growth and success are not one in the same.In an interview with Vanity Fair in 2014, Kara Swisher asked Kalanick about the nature of the interactions with the leaders of the cities (some of whom he described as “really awesome, but most are uninspired”) his business was disrupting.“If you don’t agree with the core principles, which are the premise of that compromise, then you have to have what I call principled confrontation. And so that is the thing that we do that I think can rub some people the wrong way.”If you’re beginning with the premise that compromise doesn’t work for you, and every interaction is a war of attrition, you can let people build all they want, but what exactly are you working toward?If you’re always hustlin’ — though if they do keep that one, I would urge them to restore the “g” to its rightful place — you don’t stop to think about where the pitfalls may be or whether you might be in the wrong. Uber’s experience shows us that is kind of approach is a recipe for collapsing under the weight of your own hubris.Related: If Elon Musk Can Admit His Mistakes, Then So Can YouIn Uber’s San Francisco headquarters, up until this week, the office’s main conference room was called aptly, the War Room. Bloomberg reported that along with the multitude of changes recommended by Covington & Burling, the room is now going to be called the Peace Room. It’s a bit on nose, but you can appreciate the thought, as long as it’s not just lip service.Uber’s culture is not going to change overnight, but now the company seems to have the self-awareness and tools in place to build an environment where values aren’t used, as per Holder and Albarrán’s description, to “justify poor behavior.”So what can we learn from Uber’s newfound emphasis on internal, rather than external growth? Company culture isn’t about perks or empty aphorisms that look good on a T-shirt. Simply, people want to be heard, they want their work to be valued and they want to be treated with respect. When you build your company culture, start there. Your reaction to particularly awful days can make or break future success. The better action to take is to remain as calm as possible. Take quiet time to analyze how far you have come. Think about the twists and turns, and what currently has you stopped in your tracks.Numerous conversations revealed that many almost quit their current endeavors. But one common factor as to why they did not quit helped them to prevail: They enjoy what they do too much! Ironically, it has become their way of life.As uncertainty hits, follow these two suggestions:Take the time needed to uncover the better route.Convert a negative frame of mind into a positive.Reframe the situationOne sales technique referred to as “Reframing” applies very well to difficult situations. Whether personal or trying to work with a client, the first step is to figure out where the discontent originates. Upon recognizing the misstep, it becomes easier to work together to find the better solution. By asking a series of questions such as, “What if…” the improved path begins to reveal itself.On the personal side, moving from negative to positive thought is the beginning step for improvement. The change in attitude makes an enormous difference in providing increased opportunity to save the day.The day that you find you are doubting yourself, take private time to consider “the why” that is. Most likely, one of your peers will have had a similar experience. Have a conversation about how they dealt with it and ask for recommendations on how to handle. (Editor)Mar 20 2019Next-generation sequencing of tumor DNA from patients with colorectal cancer revealed genetic alterations that were linked to different survival and treatment outcomes in an analysis led by a University of North Carolina Lineberger Comprehensive Cancer Center researcher.The findings, published in the Journal of Clinical Oncology, could help define strategies to more effectively treat colorectal cancer, the second leading cause of cancer death in the United States.”This is an example of precision oncology, where using genetics, we are able to stratify tumor types that we once believed were homogeneous, and to identify new patient subgroups that might benefit from tailored therapies,” said UNC Lineberger’s Federico Innocenti, MD, PhD, associate professor in the UNC Eshelman School of Pharmacy Division of Pharmacotherapy and Experimental Therapeutics.For the study, researchers analyzed mutations in tumors of 843 patients who participated in a phase III clinical trial. The trial compared treatment with chemotherapy plus either bevacizumab or cetuximab – regimens which are now the standard of care for this disease in the advanced stage. The researchers used tumor samples to analyze genetic mutations in the DNA. The level of tumor mutational burden defines a new subgroup of patients with better prognosis, Innocenti reported.Studies are ongoing in Innocenti’s lab to try to understand the role of the body’s immune system in contributing to the finding. Researchers want to know if these patients with higher tumor mutational burden will be more responsive to treatments that work by unlocking the immune system against cancer.”It is crucial to define which patients could be responsive to immunotherapy in this setting, and this study shows the first promising evidence to do so,” Innocenti said.The study also confirmed findings previously made for patients who had mutations in the BRAF gene in their tumors. They found these patients had worse survival compared to patients who didn’t. Median survival for patients with the mutation was 13.5 months, compared to 30.6 months in patients without mutations.”This mutation was a very strong negative prognostic factor,” Innocenti said. “There was a clear difference in survival.”There was no difference in the study between the different treatment strategies studied in the trial for patients with or without the BRAF mutation. However, Innocenti said the finding confirms these tumors have a different biology and should be treated differently.Additional studies are needed to evaluate and confirm findings with therapeutic implications in patients, Innocenti said. Further research is planned to study additional ways of stratifying patients based on their genetic features to improve outcomes. However, when their concentrations become too high, inflammation and tissue damage can occur. University of Illinois engineers have developed and tested a new drug-delivery system that senses high oxidant levels and responds by administering just the right amount of antioxidant to restore this delicate balance.The findings are published in the journal Small.Many pharmaceuticals include specialized polymers and particles that control the timing or concentration of the drug released once administered, the researchers said. However, these additives can hamper crystallization during the manufacturing phase of some drugs – like antioxidants – causing them to dissolve in the body in an uncontrolled manner.”We saw an opportunity here to develop a different kind of drug-delivery system that could sense the level of oxidant in a system and respond by administering antioxidant as needed,” said chemical and biomolecular engineering professor and study co-author Hyunjoon Kong.Kong and his team found a way to assemble crystals of catechin – the bright green antioxidant found in green tea – using a polymer that can sense when oxidant concentrations become too high. They found that the daphnids’ mean heart rates dropped from 348 to 290 and 277 beats per minute, depending on the concentration of hydrogen peroxide used.When the team added the new catechin crystal assembled with polymer to the experiment, the water fleas eventually recovered a close-to-normal heart rate.Beyond the potential pharmaceutical uses for the new polymer, Kong’s group is looking into its use for curtailing the impact of highly oxidizing chemicals in natural waterways.”Hydrogen peroxide is often used to clean water fouled by excessive algae, and this raises concern about how the oxidant may be affecting living organisms in water,” he said. “We think this new antioxidant-delivery system could be used to address the problem of over-oxidized natural waters.”The researchers plan to push ahead with developing the polymer for pharmaceutical and environmental uses. “This study proved a concept, but we have more work to do,” Kong said. “There is concern over the safety of the specific polymer we used – polyethylenimine diselenide – but we are getting close to finding a viable replacement.”
